NAD+ vs NMN vs NR: What Is the Difference?

NAD+, NMN and NR appear side by side in search results, and they are often treated as if they were the same thing. They are related, but they are three different molecules. This guide explains how they fit together.
NAD+: the coenzyme itself
NAD+ (nicotinamide adenine dinucleotide) is a coenzyme found in every living cell. It carries electrons in redox reactions, which makes it central to energy metabolism, and it is also used up by enzymes such as sirtuins and PARPs. Its molar mass is about 663 g/mol, and as the name says it is made of two nucleotides joined together.
NMN and NR: precursors
Cells keep their NAD+ supply topped up by building it from smaller molecules, called precursors:
- NR (nicotinamide riboside) is a form of vitamin B3. Inside the cell, enzymes called NR kinases add a phosphate group to turn it into NMN.
- NMN (nicotinamide mononucleotide) is one step closer. The enzyme NMNAT joins NMN with a second nucleotide to make NAD+.
- Nicotinamide (also called niacinamide) is recycled back into NMN by the enzyme NAMPT in what is known as the salvage pathway.
- Niacin (nicotinic acid) feeds a separate route, the Preiss–Handler pathway.
So the order is roughly NR → NMN → NAD+. NR and NMN are not "better" or "worse" versions of NAD+; they are earlier steps in the same chain.
NAD+ vs NMN at a glance
- Size: NAD+ is roughly twice the size of NMN (about 663 g/mol vs about 334 g/mol).
- Role: NAD+ is the active coenzyme; NMN is a building block cells convert into NAD+.
- Research focus: studies on NMN and NR usually ask whether giving a precursor raises NAD+ levels in tissues. Studies with NAD+ itself look at the coenzyme directly.
What to check on a research listing
Because the names are so similar, the most common mistake is ordering or documenting the wrong compound. Check that the label and the certificate of analysis name the same molecule (NAD+, not NMN or NR), the same strength (for example 1000mg, not 500mg), and the same batch number. Store lyophilised NAD+ refrigerated and away from light.
